Outline of Annual Research Achievements

社会性行動に必要な“愛情ホルモン” オキシトシン(OT)が中枢神経作用を発揮するには、末梢循環血中から血液脳関門(blood-brain barrier, BBB)を通過し、脳内へ移行することが必須となる。本研究では、OTの脳内移行とその作用発揮の分子機構の解明とその応用を目的として研究を推進した。その結果、BBBを構成する脳血管内皮細胞上に存在するRAGE(receptor for advanced glycation end-products)とOTとの直接結合を発見し、RAGE依存的なOTのBBB通過を確認した。続いて、合成OTをマウスに皮下注射、静脈内投与、鼻腔投与を行って解析した結果、脳血管内皮細胞に存在するRAGEがOTの脳内移行に必要であることが分かった。さらに、各種RAGE遺伝子改変マウスを駆使して検証した結果、母親マウスの子育て養育行動や末梢からの合成OT注射後の機能的神経活動も脳血管内皮細胞RAGEが担っていることが明らかとなった。また、マウス両側総頚動脈結紮モデルで一過性の脳虚血を起こし、脳血管内皮細胞のRAGE発現を上昇させると、BBBの破綻は生じないが、OTの脳内移行が亢進していた。膜型RAGEに対してデコイ受容体として機能する可溶型RAGEは、OTの脳内輸送に影響を与えず、むしろ輸送を高める傾向があった。別の実験として、新しいOT結合タンパクを同定するため、金属ビーズにOTを化学結合したもので血清中の結合キャリアタンパク候補を質量分析で複数同定し、そのノックアウトマウスを作製し、現在解析を行っている。
血中OTが血液脳関門(BBB)を通過し脳内へ移行する際の機能分子RAGEが初めて同定され、OT研究が進展した。
