Outline of Annual Research Achievements

近年の癌ゲノム研究によって分子サブタイプ分類が進み、ゲノム医療・プレシジョンメディシンへの応用が期待されている。消化器癌においてもARID2変異肝癌、KDM6A/TP53変異膵癌、CDH1/RHOA変異胃癌など難治性サブタイプの存在が明らかになったが、その分子生物学的意義が不明なため治療標的の開発には至っていない。我々はゲノム編集技術を用いてARID2変異ヒト肝癌を解析し、DNA修復異常によってhypermutatorとなることを証明した(J Hepatol 2017)。本研究では、多重ゲノム編集を可能とする新規レンチウイルスベクターの開発を目的とする。癌細胞には多くの遺伝子変異が蓄積しており、多重ゲノム編集技術によって分子サブタイプの解明のみならず、同系マウスがん細胞パネルの構築が可能となり、免疫チェックポイント阻害剤との併用療法など新規治療開発へも応用できる。実臨床を目指したプレシジョンメディシン実装化のブレークスルーとなる挑戦的かつ戦略的研究課題である。
本年度は、multiplex system と LentiGuide systemの各々の長所を取り入れて、３遺伝子を標的とする新たな多重ゲノム編集ウイルス LentiGuide-multiplex systemの構築に成功した。DEPDC5(Sci Rep 2018)、DAXX(Endocr Relat Cancer 2018)、KDM6A(Int J Cancer 2019)、PRMT6(Carcinogenesis 2019)のゲノム編集解析により、分子生物学的、臨床病理学的意義を解明した。さらにイントロンの2重ゲノム編集によって、エキソンスキッピングの高効率誘導にも成功しており、計画以上の成果が得られている。現在、正常免疫マウスにおいて免疫抵抗性クローンの作製を進め、治療標的の同定と解析を加速させている。

Reason

多重ゲノム編集には複数のsgRNAを発現するプラスミドやアデノ随伴ウイルスを用いた報告が多いが、導入効率やサイズ制限などの問題を克服するため、簡便なレンチウイルス・システムの開発が必須である。Charles Gersbachラボからmultiplex primer setsを搭載したone-vector systemが報告されたが(Nucleic Acid Res 2014)、複雑なデザインでサイズも大きく、変異導入効率が低いため 一般化されていない。一方、Feng Zhangラボが開発したLentiGuide systemは単一ゲノム編集用であるが、one-vector systemに比べて数十倍効率が高く、その簡便さから広く普及している(Nat Methods 2014)。今年度の挑戦的研究（萌芽）によって、multiplex system (Gersbach)と LentiGuide system (Zhang)の各々の長所を取り入れた、３遺伝子を標的とする新たな多重ゲノム編集ウイルス LentiGuide-multiplex systemの開発に加え、イントロンの2重ゲノム編集によってエキソンスキッピングの高効率誘導にも成功し、計画以上の成果が得られている。

Strategy for Future Research Activity

現在、正常免疫マウスにおいて免疫抵抗性クローンを作製し、治療標的の同定と解析へと進展している。癌は多様な遺伝子異常の多段階蓄積によって成立している。簡便かつ迅速な多重ゲノム編集技術を確立にすることができれば、分子サブタイプの多様性に応じた免疫システムの解析が可能となる。本年度の成果により、ノックアウトとエキソンスキッピングを組み合わせた分子サブタイプの再現が加速化されており、正常免疫状態における各種がんパネルの構築を進める。がん臨床において、免疫チェックポイント阻害剤の単剤効果は20-30％程度にとどまっており、分子標的治療剤などとの併用治療への期待が高まっている(Sharma & Allison. Cell 2015)。正常免疫状態における腫瘍微小環境の詳細な解析によって、各サブタイプ特異的な免疫複合療法の確立を目指している。

Causes of Carryover

当初の計画より多重ゲノム編集ウイルス作成が早く進み、試薬および解析費用の節約となった。論文投稿は順調に進んでいるが、アクセプトされた論文の一部が平成30年度末の段階でin pressであったため、論文関連費が繰り越しとなっている。
