活動依存的カルシウム流入による競合的シナプス回路発達の共通原理の解明

Research Abstract

NMDA受容体のGluN2BとGluN2Dは発達段階に発現する2つのサブユニットである。本研究では、この2つのサブユニットの機能的差異について、GluN2BヘテロマウスとGluN2Dホモ欠損マウスの体性感覚系の回路発達を比較した。同腹のコントロールマウスでは、三叉神経核脊髄路核のバレレットは出生日(PO)に、視床のバレロイドはP2-P3、体性感覚野のバレルはP4-P5に出現した。これに比べ、GluN2Bヘテロマウスではそれぞれ1日遅れて出現し、GluN2Dホモ欠損マウスでは1日早まった。片側眼窩下神経切断による臨界期終了時期を検討したところ、GluN2Bヘテロマウスでは1日遅れて終了し、GluN2Dホモ欠損マウスでは1日早まった。したがって、体性感覚路全般に渡って、GluN2Bはシナプス回路の形成と改築を促進し、対照的にGluN2Dはこれを遅延させていることが判明した。転写レベルでは、GluN2Bは前脳にGluN2Dは脳幹に豊富に発現することが判っている。特異抗体を作成してそのニューロン発現とシナプス局在を検討したところ、成体期の三叉神経核・視床・体性感覚野のいずれにおいても、GluN2Bは主にprincipal neuronに発現し、その非対称性シナプスのシナプス後部に局在した。これに対して、GluN2DはGABA陽性ニューロンにほぼ選択的に発現し、その非対称性シナプスのシナプス後部に局在していた。同様の細胞発現パターンは、生後第1週においても観察されたことから、GluN2Bはprincipal neuronに発現してそれが作る興奮性シナプス回路発達を促進し、GluN2Dは抑制性ニューロンに発現して興奮性ニューロンによるシナプス回路発達を遅延させると結論した。
