拒食症の感受性遺伝子の網羅的同定と機能解析による発症カスケードの解明

Research Abstract

CNTN5、SYT12、NETO1、PARK7は、独自のゲノムワイドマイクロサテライト相関解析の結果同定した摂食障害感受性遺伝子であり、何れもが神経系で機能すると推測されている。SYT12、NETO1についてはENU点突然変異マウスを計5個同定し、スピードコンジェニック法を用いた純化を行っている。先に取得したCNTN5 KOマウスも含め、これらの変異体マウスは、通常の飼育条件下では明らかな摂食行動の異常を示さない。より詳細な行動解析・神経機能解析が必要であり、摂食に伴いストレス負荷をかける等、摂食障害の発症機序をより反映する条件での解析を準備している。また、これら遺伝子のネットワーク解析を目的とした酵母ツーハイブリッド解析を進行中である。従来法による解析の結果得られた相互作用因子情報は極めて限られていたため、従来法の10倍程度の検出力を持つ新たな酵母ツーハイブリッド法を独自に開発し、これを用いて相互作用因子解析を続行している。
