拒食症の感受性遺伝子の網羅的同定と機能解析による発症カスケードの解明

Research Abstract

CNTN5、SYT12、NETO1、PARK7は、我々独自のゲノムワイドマイクロサテライト相関解析を用いて、新規に同定した摂食障害感受性遺伝子であり、何れもが神経系で機能すると推測される。本年度については、これら摂食障害感受性遺伝子のなかで既に作成したSYT12とNET1についてはENU点突然変異マウス計5種類の遺伝子改変マウス、並びにCNTN5ノックアウトマウスについて引き続き行動解析を進めたが、いまのところ摂食障害に関連するような表現型は観察されていない。
一方、これらの摂食障害感受性遺伝産物について、相互作用ネットワーク解析を効率よく進めるため、従来は原理的に不可能であった「ランダムcDNA同士の無作為的・網羅的相互作用解析」を可能にする高効率・高精度の新規酵母Two Hybrid(YTH)法を開発した。この新規YTH法は従来法の10倍の検出力を有することが明らかとなった。そこでこの新規YTH法を用いて、摂食障害感受性遺伝子SYT12とNETO1を中心として相互作用解析を行ったところ、シナプス開口放出・リサイクリングに関与する多数の因子と相互作用することが見出された。さらに、免疫沈降法によって相互作用の検証を行い、SYT12やNETOがシナプス小胞開口放出やリサイクリングに関与する因子群と相互作用することを実証するとともに、それ自身の相互作用を生化学的に確認した。
これらの知見をもとに、摂食障害感受性遺伝子を基盤とする創薬への糸口として、我々が見出した相互作用を阻害するペプチドや低分子の同定を進めている。
