Outline of Annual Research Achievements

子宮内膜上皮のプロゲステロン(P4)受容体(PGR)欠損マウスでは着床期の子宮内膜上皮の細胞増殖抑制が起こらず、上皮でも間質でもP4応答能が低下し、着床に必須のサイトカインLIFの発現が低下して胚接着が障害された。LIFのシグナル調節因子STAT3に着目し、子宮内膜上皮、子宮内膜間質、子宮全体のSTAT3欠損マウスを作成した。上皮と間質のSTAT3欠損マウスはいずれも胚接着が障害された。上皮のSTAT3欠損マウスは子宮管腔が入り組んだ構造を示し、間質のSTAT3欠損マウスは上皮のエストロゲン応答能亢進と持続的な細胞増殖を示した。上皮のSTAT3は着床期の子宮管腔のスリット状形態変化を制御し、間質のSTAT3は上皮のエストロゲン応答能と細胞増殖能を抑制することが示され、上皮と間質のSTAT3が異なる経路を介して胚接着を制御することが分かった。まとめると、本研究により、プロゲステロン受容体、STAT3が着床期の子宮内膜上皮の増殖抑制に関与することが示された。
