Outline of Annual Research Achievements

ヘリコバクター・ピロリ (ピロリ菌)は全人類の約半数が感染し、胃癌を始めとする多彩な疾患を引き起こす世界最大の感染症の一つであるが、未だ解決していない「問い」がある。1) 一部のピロリ菌感染者だけが疾患を発症し、しかも人によって発症する疾患に違いがでるのはなぜだろうか？ 2) 胃癌発症率に 地域差が生じるのはなぜだろうか？ これらの「問い」に答える手段として、我々は、オルガノイド(胃ではガストロイド)を2次元化し、さらに上皮下組織の一部再現モデルとして、2次元化したガストロイドとT細胞を共培養する系を米国にて確立した。本研究では、既知の病原因子の真の機能の探求、さらに、我々が保有する世界最大規模数のピロリ菌を用いて、菌の全ゲノム解析を行い、疾患の発症に関与する新規病原因子の発見を目指す研究を展開する。
最終年度は、引き続くコロナ禍のため米国との直接的な現地での共同研究を進めることができなかったため、最終的に2次元化したガストロイドとT細胞を共培養する系を大分大学で確立することはできなかった。しかし、本学において独自に日本人を用いたオルガノイド作成を進め、各種ピロリ菌および変異株との共感染モデルを確立して研究を進めた。
また、新規病原因子の探求に関しては、次世代シーケンサーを駆使した解析を精力的に行い、バイオフィルムに関与する遺伝子、さらにファージやプラスミドに病原性があることなども発見し、ピロリ菌に関して多くの論文を発表することができたことは大きな成果である。また、本事業は、新たに獲得した基盤研究（B）にて継続可能で、さらなる成果が期待できる。
