Outline of Annual Research Achievements |
今年度は,1細胞ショットガンプロテオーム解析を実施するためのシステム統合を検討した.具体的な操作方法としては,半自動ナノピペットデバイスを使用して細胞サンプリングキャピラリー内に細胞溶解バッファー,ターゲット細胞,細胞溶解バッファー,トリプシン溶液の順に顕微鏡下で連続的に吸引した.次に,細胞サンプリングキャピラリーを50 ℃のオーブン内で30分間反応させることにより,ペプチド消化物を得た.この新たな1細胞プロテオーム解析のための試料調製法 (in-line sample preparation for efficient cellular proteomics, ISPEC-P 2.0) は従来のISPEC-P 1.0と比較して1細胞のサンプル調製にかかる時間が2時間から1時間に短縮された.次にISPEC-P 2.0の実用性について,カラム内径のダウンサイジング(内径30μm)と 密閉型Nano-ESIイオンソースを用いた高感度Nano-LC/MS/MSプロテオーム分析システムを用いて評価した.その結果,HeLa1細胞から421ペプチド,166タンパク質の同定に成功した. 最後に,開発した超高感度解析システムを用いて,細胞周期の進行をリアルタイムに観察可能なFucci (Fluorescent Ubiquitination-based Cell Cycle Indicator) HeLa 細胞の細胞周期ごとのタンパク質の情報の取得を行った.顕微鏡下で蛍光を指標にG1、S、G2/M期の細胞を各10個ずつキャピラリー内に回収処理しショットガンプロテオーム解析による比較を行った.その結果G1期,S期,G2/M期からそれぞれ460,396,564種類のタンパク質を同定することに成功した.
