Outline of Annual Research Achievements

令和3年度はまず、Cyclopoida目に属するカイアシの一種を対象とした各種多価不飽和脂肪酸(PUFA)生合成酵素遺伝子の単離およびその機能解析を進めた。まず、当該種の高精度トランスクリプトームデータベースを構築し、相同性検索によりPUFA生合成に関わる各種不飽和化酵素遺伝子および鎖長延長酵素遺伝子配列の網羅的単離を行った。続いて、これら遺伝子配列を出芽酵母に発現させることで、その機能を解析した。その結果、本種は前年度までに解析したHarpacticoida目カイアシであるTigriopus californicusやPlatychelipus littoralisとは異なるPUFA生合成経路を有することが明らかとなり、カイアシ類のPUFA生合成経路は高度に多様化していることが強く示唆された。続いて、前年度から検討を重ねていたリポソームによる安定同位体標識脂肪酸の封入および、カイアシ類への投与を行った。本手法によりT. californicusに安定同位体標識オレイン酸を取り込ませることに成功し、液体クロマトグラフィー質量分析法により安定同位体比を解析することで取り込まれたオレイン酸がEPAおよびDHAまで代謝されることを明らかにした。本結果は前年度までに解析した本種の各種PUFA生合成酵素の機能と合致するものであり、T. californicusが確実に非PUFA(オレイン酸)からDHAまでの生産能を有することが明らかとなった。すなわち、カイアシ類の中には、PUFAの一次生産者として機能し得る種が存在することが示された。なお上記成果をまとめて、令和4年度日本水産学会春季大会にて口頭発表を行った。
