Outline of Final Research Achievements

We developed the analytical system that combines calcium transient data with static immunostaining images in individual cultured cardiomyocytes obtained by high-content imaging system. We analyzed calcium transient data in murine cultured cardiomyocytes or in human induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) and developed MATLAB program that links the obtained data. Finally, we developed sarcomere live-imaging system in iPSC-CMs using genome editing.

Academic Significance and Societal Importance of the Research Achievements

カルシウムは、心筋細胞において、興奮収縮連関の中心的役割を担う。本研究では、個々の心筋細胞レベルにおけるカルシウムの動態を、免疫染色で得られる画像情報と1対1でリンクさせ、自動解析するアルゴリズムを開発した。更に、ヒトiPS細胞由来分化心筋細胞を用いたサルコメアライブイメージング法を確立した。ゲノム編集技術や、疾患iPS細胞技術と組み合わせることで、遺伝性心筋症研究への応用が期待できる。
