Outline of Final Research Achievements |
Monoclonal antibodies showing different reactivity to beef, pork and chicken myoglobin (Mb) were obtained. The obtained antibodies were used to construct a sandwich ELISA, which is a more practical pork determination method compared to conventional methods. This ELISA did not cross-react with beef Mb, chicken Mb, lamb, or goat meat, and was able to quantitatively measure pork Mb and detect 1% (w/w) pork added to beef. It was also able to detect pork with similar sensitivity when cooked after pork addition. Furthermore, the obtained and commercially available antibodies were applied to a surface plasmon resonance (SPR) measurement instrument to construct an SPR immunosensor. It was suggested that this immunosensor was capable of simultaneously quantifying beef, pork, and chicken.

Academic Significance and Societal Importance of the Research Achievements |
従来、食肉偽装対策や原材料表示を監視するために食品中の豚肉量を測定するイムノアッセイがいくつか開発されているが、食品の調理・加工によるタンパク質の変性や抽出効率の低下の影響を受けるため、正確な測定が困難だった。本研究で構築したELISAはこの問題を克服し、生肉及び加熱調理肉中の豚肉を正確に定量でき、食肉製品の認証や誤表示の防止への利用が期待できる。さらにイムノセンサ構築を含め得られた成果は、食肉の分析に限らず、他のアレルギー原因食品の分析にも応用可能である。将来的に特定原材料の一斉分析法開発への道を拓くと共に、食肉に関する宗教や信条により課せられる食品選別法の開発に役立つ知見を提供できる。
