神経可塑性及び脳の発達におけるIP_3受容体のカルシウムシグナリングの解析

Research Abstract

IP_3受容体タイプ2型、3型ノックアウトマウスが外分泌障害や味覚障害を起こすことをすでに報告したが、更に小脳の顆粒細胞の分化を促進する働きを持つことを証明した(Futatsugi,J.Neurochem.2008)。また、タイプ2型、3型のノックアウトマウスに関して嗅覚器の組織学的な解析を行ったところ、鼻粘液分泌の顕著な低下が見られ、生体内イメージングや行動テストにより嗅覚機能の低下を確認した(Fukuda,Eur.J.Neurosci.2008)。次に、IP_3受容体タイプ2型をノックアウトした細胞が破骨細胞の単独培養法では分化が著しく阻害されることに注目し骨細胞の分化誘導の比較を行った。その結果、RANKL添加によって観察されるカルシウムオシレーションにはIP_3R type2が必須であること、カルシウム/カルシニューリン/NFATc1というシグナル経路が阻害されると破骨細胞の分化が妨げられることを明らかにした(Kuroda,PNAS 2008)。更に、yeast two hybrid法よりIP_3結合蛋白質であると同定されたプロテインキナーゼC基質80K-Hが新規のIP_3受容体のレギュレーターであることを証明した(Kawaai,JBC 2009)。次に、小脳のPurkinje細胞のシナプス後部に特異的に存在するHomer3に着目し、Homer3が神経活動に依存してリン酸化され、それによりHomer3と代謝性グルタミン酸受容体との複合体が消失することを発見した(Mizutani,J.Neurosci.2008)。また、カルシウムストアが発達中の背根神経節神経細胞の成長円錐においてマクロピノサイトーシスを誘導する事を初めて明らかとし、マクロピノサイトーシスによる大規模な細胞膜の回収が反発性軸索誘導のメカニズムである可能性を示した(Kabayama,Mol Cell Neurosci 2009)。
