Research Abstract |
細胞内で起こる生命現象の多くは、複数の因子が特定の時期に特定の場所に集合し、機能を発揮する反応である。遺伝情報を担う染色体DNAの複製においても、まずDNAを合成する酵素を含む多数のタンパク質が複製を開始するDNA領域に集合し、次にこれらタンパク質群が何らかの変換(リモデリング)をし、DNA合成を始める。本研究では、出芽酵母の染色体DNA複製開始過程をリモデリングのモデルとし、タンパク質の複製開始領域への集合とその後に起こるタンパク質複合体の変換(リモデリング)の分子機構、そして細胞周期によるこれらの調節機構の解明を目指す。そのため、複製開始領域へ集合するタンパク質群の挙動を細胞内で調べるとともに、これらタンパク質を精製し、タンパク質間の相互作用・複合体形成や、開始領域への集合を試験管内で詳細に調べる。これまでに、pre-LC複合体、Sld3-Sld7複合体を新たに同定し、これら複合体が複製開始領域への集合反応とその後のリモデリング反応に働くことを示すことができた。
