Research Abstract

糖・脂質・エネルギー代謝は生命発現にとって極めて重要である。インスリン(Ins)とアディポネクチン(Ad)はこれら代謝制御の2大経路である。我国で死因上位の心血管疾患の主因である糖尿病・メタボリックシンドローム(MS)の激増の原因解明と治療法開発には、代謝制御とその破綻の統合的理解が不可欠である。本研究では、各種各組織遺伝子欠損マウスを駆使し、中枢・末梢の各組織及び全身におけるAd作用とIns作用の全容の解明を中心にして、代謝制御機構の統合的理解とその破綻の改善法開発に至るまでの(1) 代謝制御における臓器間クロストークのメカニズム解明及び(2) 代謝制御における細胞機能・恒常性のメカニズムの解明を目的とした。

骨髄移植と血管内皮特異的遺伝子欠損の解析により、AdipoR1、R2それぞれの抗動脈硬化作用における役割と細胞ないシグナルを明らかにした(未発表)。また、中枢のIRS-2が摂食、肥満、レプチン感受性と肝臓でのIns感受性調節に重要な役割を果たしていることを明らかにし、中枢ー肝臓間クロストークのメカニズムを明らかにした(未発表)。また、Insシグナルの鍵分子であるAktの骨格筋特異的欠損マウスを解析し、骨格筋におけるAktシグナルが加齢に伴う筋肉量の減少に関与し、Ins抵抗性の原因の一部を説明できることを示した(未発表)。

肝臓特異的R1/R2欠損の解析により、肝臓ではR1が糖新生抑制、R2が糖取込における役割分担を明らかにし、それぞれの細胞内シグナルを解析した(未発表)。また、網羅的発現解析により、肝臓での小胞体ストレス調節転写因子XBP-1sとその下流分子Sdf2l1の病態生理的意義を明らかにし、さらにその発現を反映する発光マウスを作製し、健常状態と肥満状態での発光が変動し、生理的な小胞体ストレスの阻害、Ins抵抗性に関連する可能性を示した(未発表)。
