APP小胞輸送機構の異常とアルツハイマー病発症機構

Research Abstract

アルツハイマー病(AD)の有力な発症原因はβアミロイド(Aβ)の生成とオリゴマー形成による神経毒性の発現である。Aβは前駆体タンパク質APPが2回の連続的切断を受けて生成する。毒性の強いAβが生成・分泌される仕組みを解明する事は、ADの根本治療法を開発する上で不可欠である。APPは、JIP1タンパク質を介してキネシン-1と結合する膜小胞カーゴ受容体であり、神経細胞体から末端にかけて順行性のモーター輸送を受ける。輸送過程が何らかの障害を受け、APP小胞輸送が滞る、APPのアミロイド生成的代謝が亢進しAβの生成が増加する。本研究では、APPがJIP1を介してキネシン-1の軽鎖(KLC)に結合する分子機構を解明し、APPとモーター分子の結合解離を制御している制御機構の解明に取り組んだ。まず、APP-JIP1およびJIP1-KLCのどちらの結合制御機構がAPP小胞輸送の調節に関わっているかを解明し、JIP1-KLCの貢献度が高いことを見いだした。次にJIP1-KLCの結合を制御する領域の同定を両タンパク質の変異体を細胞に発現させ行った。同定した領域の制御機構として、タンパク質リン酸化系を見いだした。細胞におけるJIP1およびKLCの複数のリン酸化サイトを決定し、in vivoで機能しているサイトの同定に取り組んでいる。AD脳では、タウタンパク質のリン酸化亢進を始めとしたタンパク質リン酸化・脱リン酸化系の異常が報告されており、老化脳におけるJIP1およびKLCのリン酸化制御異常が、APPの小胞輸送に影響を与え、APPをアミロイド生成的代謝に向かわせている可能性が考えられた。
