Research Abstract

生殖細胞の形成過程には、通常の体細胞とは異なる細胞の増殖、成長が観察される。本研究では、ショウジョウバエの生殖系列細胞に特異的な細胞増殖と成長を制御する機構を制御する機構を明らかにする研究を進めてきた。今年度は、とくに精母細胞の成長を誘導する機構を解析した。減数分裂開始前の成長期において精母細胞は容積を25倍増加させる。これまでにインスリン様ペプチドの産生細胞を欠く個体およびインスリン受容体の突然変異体では精母細胞の成長が阻害されることをみいだした。そこでこの細胞成長を誘導するシグナル伝達因子の遺伝学的同定を試みた。同受容体の下流因子IRS/Chicoの機能欠損型突然変異体では精母細胞の有意な成長阻害が見られた。一方、シグナル伝達因子PI3キナーゼ、RAS87Dの活性型変異Dp110-CAAX、RAS85D^<v16>をそれぞれ精母細胞で発現させると、精母細胞の過度な成長がみとめられた。この結果は精母細胞の成長を促すインスリン様ペプチドのシグナルはRas/MAPK経路およびPI3キナーゼ/Akt経路の両方を介して伝達されることを示唆している。Aktは細胞成長を負に制御する転写因子Foxoをリン酸化することにより抑制する。Aktリン酸化サイトを改変したFoxo変異体はAktによる抑制を受けない。この変異体を発現させたところ精母細胞への影響は認められなかった。Foxoは精子幹細胞の分裂には負の調節因子として働くが、精母細胞の成長には関与しない可能性が考えられる。さらにインスリン経路と平行するTorシグナル伝達経路の関与を検討するため、Tor機能欠損型変異、その活性化因子Rhebならびに抑制因子Tsc1,Tsc2の機能欠損変異体において精母細胞の成長を調べた。その結果、これらの制御因子を介したTorシグナル伝達系も精母細胞の成長誘導に関与することが強く示唆された。
