| Research Abstract |
クモ膜下出血後の脳血管攣縮発生には血管平滑筋細胞内にある蛋白リン酸化酵素C(PKC)が重要な働きをしており、中でもPKCのisoformであるPKCδとPKCαがその発生、維持に大きく関係していることがわかっている。これをふまえPKCδとPKCαの両者の阻害剤であるM-62885という物質を至適濃度でクモ膜下出血後の脳内に投与できれば脳血管攣縮は抑制されると考えられる。そこで治療における至適濃度はどのくらいか、副作用はどうかを明らかにする。その後この薬剤が徐放性にクモ膜下腔に放出されるようなpelletを開発し、その中にこの薬剤を注入し、クモ膜下出血後に脳層内に設置して脳血管攣縮発生が有効に抑制することができるかを検証する。
