| Outline of Annual Research Achievements |
1. 心筋機能低下におけるCaMK-NRSF-GalphaO circuitの意義の解明。昨年度までに、NRSF心筋特異的ノックアウトマウス、優性抑制変異NRSF過剰発現マウスを用いた解析で心機能低下におけるGNAO1発現亢進の病的意義を新規に見出し、GNAO1ノックアウトマウスやGNAO1心筋特異的過剰発現マウスを用いてその意義と分子機序を解明し、成果をCirc Res誌（IF:23.2）に発表した（Inazumi H, et al. Circ Res 130:234-248.2022）本年度は、GNAO1遺伝子改変マウスの解析によるGNAO1の下流シグナル探索を行うと共に、GNAO1を標的とした新規核酸医薬の開発に着手した。
2. ヒストン脱アセチル化酵素HDAC1/2を介したCaMK-NRSF転写経路とNotch-Hrt2転写経路のクロストークとその心筋リモデリングにおける意義の解明。心筋特異的誘導性HDAC1/2、NRSF 、Hrt2それぞれの心筋特異的誘導性ノックアウトマウスの心臓の表現型および遺伝子発現解析、エピゲノム解析を継続した。
3.受容体活性化型カルシウムチャネルTRPC3/6の新規病的心筋リモデリング予防・治療標的としての意義の解明。複数の心不全モデルにおいてTRPC6、TRPC3ノックアウト、およびTRPC3/6阻害薬の効果の解析を継続した。また共同研究にてTRPC6活性化がβアドレナリンシグナル増強を介して急性心不全時の心機能維持に寄与することを見出し、Nature communicationに発表した（Oda S, et al . Nat Commun.13(1):6374.2022）。
4. LSD1、G9aの心筋恒常性維持における意義の解明。LSD1およびG9aの心筋特異的ノックアウトマウスの表現型解析、遺伝子発現解析を継続した。
