Outline of Annual Research Achievements

緑内障は視野欠損が不可逆的に進行することによって失明に至る眼疾患である。落屑症候群から発症する緑内障の病型の一つである落屑緑内障については、眼組織を構成するエラスチンの重合酵素をコードする遺伝子(LOXL1)上のゲノム配列の違い(バリアント)が疾患に関連することが報告されている。しかし、疾患のリスクを規定するバリア ントのアレルが日本人と白人とで逆転していることが判明したことから、LOXL1以外の関連分子の存在が強く示唆されている。そこで本研究では、日本人検体を 用いたマルチオミクスを実施し、LOXL1周辺領域の配列情報や転写産物を精査することによって、落屑緑内障の発症機序を明らかにすることを目的とする。
最終年度には、昨年度までに実施した落屑症候群／落屑緑内障の次世代シーケンサーを用いたゲノム解析（ゲノミクス）に加えて、トランスクリプトーム解析（トランスクリプトミクス）に向けた眼組織（前嚢、虹彩、線維柱帯）の収集と凍結破砕器（SK mill）を用いたtotal RNAの抽出方法の最適化を行い、トランスクリプトミクスに向けた基盤整備を完了した。今後、国際大型共同研究の枠組みの中で、日本人を含むアジア人と白人由来のゲノミクス／トランスクリプトミクスデータについて、LOXL1とその近傍遺伝子（TBC1D21やPML）だけでなく、エキソーム解析により新たに同定したCYP39A1の配列情報や転写産物を詳細に比較することにより、人種差に基づいた落屑緑内障の発症機序の解明に至るのではないかと考えている。
