統合失調症の発症・病態仮説に共通するトリプトファン代謝に注目した予防・治療戦略

Outline of Annual Research Achievements

妊娠期に母親がインフルエンザに感染すると統合失調症に対する発症脆弱性が形成されるという神経発達障害仮説に基づき、ウイルスRNA様の作用を示す合成二本鎖RNAポリイノシン-ポリシチジン(PolyI:C)を妊娠マウスに投与したところ、この母体から出生したマウスは統合失調症様の行動異常を示した。そこで、妊娠期ウイルス感染が母体や胎児脳のトリプトファン代謝にどのような影響を与えるか検討したところ、妊娠期PolyI:C投与により胎児脳において3-ヒドロキシキヌレニンが増加することを見出した。次に、妊娠期PolyI:C投与による統合失調症様の神経発達障害・行動障害に対するトリプトファン代謝に注目した病態解明およびそれを標的とした治療薬開発について、3-ヒドロキシキヌレニンを合成する酵素であるキヌレニン-3-モノオキシゲナーゼ(KMO)遺伝子欠損マウスへの妊娠期PolyI:C投与およびKMO阻害剤の併用投与により、1)妊娠期PolyI:C投与による胎児脳における大脳皮質層構造形成および神経新生・発達の異常が認められなくなったこと、2)その出生仔の統合失調症様の行動学的異常も認められなくなった。以上の結果より、母体の免疫応答によりKMOを活性化し、代謝産物の3-ヒドロキシキヌレニンの神経毒性により胎児の神経発達障害を惹起し、発症脆弱性を上昇させることが示唆された。

母体の免疫応答によりKMOを中心としたトリプトファン代謝経路の活性化すること、それが出生仔の統合失調症様の行動障害に関与することを明らかにできたことにより、当初の目標を十分に達成できているため。

Strategy for Future Research Activity

今後、母体の免疫応答により他のトリプトファン代謝酵素の発現がどのように変容し、それが出生仔の統合失調症様の行動障害に関与するか明らかにすることで、より網羅的な解析を実施する。
