Outline of Final Research Achievements |
In this study, primary cancer was surgically removed from nine colon cancer patients and cancer organoid culture was performed. Cancer organoids were established in eight of the nine cases. Genes encoding GFP and tomato fluorescent proteins were successfully introduced into cultured cancer organoids using lentivirus, and they are now ready for transplantation experiments into mice. In the future, we would like to use PDX models using GFP- and tomato-positive colon cancer organoids to examine the antitumor effects of drugs and conduct biological research on metastasis.

Academic Significance and Societal Importance of the Research Achievements |
難治性の大腸癌は、浸潤・転移や治療抵抗性を示す予後不良な癌である。しかしながらその悪性化の分子メカニズムの理解は十分ではく、有効な治療法が限られている。本研究は、患者大腸癌サンプルを用いてPDXやオルガノイドを使用した患者個別化モデルを作製し、新規治療法の開発に役立てることに社会的意義がある。
