Outline of Final Research Achievements |
The aim of this study is to elucidate that the relationship between epithelial cell-derived OPN and colitis, colitis associated cancer, and gut microbiota by using OPN bone marrow chimeric mice. It was suggested that epithelial cell-derived OPN is associated with the regulation of gut microbiota. In the acute colitis-induced model, colitis was more severe in the group in which epithelial cell-derived OPN was knockout. In the colitis associated cancer model, the group in which epithelial cell-derived OPN was knockout had a large number of tumors but a small size. These data indicate that epithelial cell-derived OPN is involved in the regulation of colitis, colitis associated cancer, and gut microbiota.

Academic Significance and Societal Importance of the Research Achievements |
潰瘍性大腸炎 (Ulcerative colitis、以下UC)は、若年者に好発する病因不明の炎症性腸疾患であり、近年本邦においてその患者数は増加の一途をたどっている。UCの原因はいまだ不明であり,現時点で根治治療は存在しない。また、長期罹患のUC患者の増加と共に、腸炎を背景としたdysplasiaや炎症性発癌が臨床上の問題となっている。我々の研究結果により、上皮細胞由来OPNが大腸炎や炎症大大腸癌ならびに腸内細菌叢の制御に関与することが示唆された。今後ヒトサンプルを用いて潰瘍性大腸炎におけるOPNの役割を更に検討することが、増加し続ける潰瘍性大腸炎患者の治療方針決定につながると期待される。
