生活習慣病の病態におけるアルドステロン/鉱質コルチコイド受容体活性化機構の解明

Research Abstract

1、細胞培養系、ならびにin vivoの酸化的ストレスによる心筋拡張障害モデルを用いて、ROSの過剰産生による心臓拡張機能障害モデル動物で鉱質コルチコイド受容体とFoxo1が相互に関連をもち、心不全関連遺伝子を活性化させる可能性が明らかになった。これらの結果を元に、心筋特異的のFoxo1欠損マウスの作成を進めた。
2、マウスの大動脈に狭窄を作ることによって心臓に対して圧力を負荷するモデルを作成し、検討を進めた。本圧負荷モデルの心臓病変形成過程にRac1-MR系が関与することを示す可能性が明らかになり、遺伝子改変動物の作成を進めた。
3、糸球体構成細胞ポトサイト特異的にRac1が過剰活性化するマウス(RhoGDIーKO)を作成し解析を進めた。その結果、本モデルマウスが著明な蛋白尿を生じることを見出した。さらに、その蛋白尿は鉱質コルチコイド受容体拮抗薬によって減少し、腎組織の障害を著明に抑制することができることを明らかにした。

Reason

昨年度までに培養系を用いての得られた結果を裏付けるように、本年度は異なる3つの動物モデルの系でも同様の結果が得られてきている。さらに、これまでに明らかにしてきたRac-MR系に加えて、従来不明であった酸化的ストレスがなぜMR系を活性化できるのかという命題について、分子生物学的な機構がin vitro、in vivoの面から明らかになりつつある。

Strategy for Future Research Activity

動物実験系として酸化的ストレスによる心筋拡張障害モデル、心臓圧負荷(TACモデル)および糸球体障害モデルの解析を進める。さらに、心筋特異的のFoxo1欠損マウス、Rac1過剰活性化動物などの遺伝子改変動物の、より詳細な解析をすすめ、酸化的ストレスによるMR活性化機構の最終的な解明を目指す。
