神経芽腫の増殖・分化・細胞死を制御する新規NLRR受容体ファミリーの解析と応用

Research Abstract

我々が作製した神経芽腫cDNAライブラリーから同定した新規ヒト依存性受容体であるNLRRファミリーついて解析を進め、以下の知見を得た。(1)NLRR1は神経芽腫の悪性度と有意に相関して発現していたが、NLRR1受容体分子自体には機能ドメインが無く、他の増殖促進シグナルを発信する膜受容体(EGFR,IGFR等)と機能的結合することにより、がん細胞の増殖を促進していることが明らかになった。そのメカニズムにlipid raftが関与している可能性が示唆されたが、今後それを証明する必要がある。ノックアウトマウスの作製に成功し、解析を始めつつある。(2)NLRR2は、細胞に過剰発現すると細胞死を誘導した。この細胞死は、短い細胞内ドメインを欠く変異体では誘導されなかったことから、この領域を介して何らかのシグナルが細胞内に伝わっているものと思われた。現在、いくつかの細胞内death signalsを検討中である。NLRR2のノックアウトコンストラクトを作製しES細胞のスクリーニングまで行ったが、再度コンストラクトを作成中である。(3)NLRR3は、神経芽腫細胞に過剰発現すると分化を誘導し、その発現を抑制すると細胞死が誘導された。分化誘導の際には、Akt,ERK1/2の活性化が見られたため、他の神経分化を誘導する膜受容体との機能的関係が示唆された。NLRR3のノックアウトコンストラクトを作製した。NLRRファミリーのアミノ酸配列はアフリカツメガエルまで非常に高く保存されており、その生理機能の解明は正常神経発生と発がんの分子機構解明に極めて有用と思われた。
