神経芽腫の増殖・分化・細胞死を制御する新規NLRR受容体ファミリーの解析と応用

Research Abstract

我々が作成した神経芽腫に特化したcDNAライブラリーから新規ヒト依存性受容体であるNLRRファミリーを同定し、その解析から以下の知見を得た。(1)NLRR1は進行神経芽腫において有意に発現が高く、なかでもMYCN増幅した神経芽腫において高発現した。そこで、NLRR1強制発現系において解析したところ、NLRR1高発現によりSP1が誘導され、SP1によるMYCNの転写誘導が見られた。また、逆にMYCNを高発現させるとNLRR1の転写が誘導され、NLRR1はMYCNの直接の転写ターゲットであることが明らかになった。したがって、悪性神経芽腫においては、NLRR1-MYCNのpositive feedback loopが存在し、増殖を促進していた。一方、NLRR1ノックアウトマウスの作成に成功し、詳細な解析を進めている。(2)NLRR2の過剰発現により細胞増殖の抑制、コロニー形成能の抑制が生じ、siRNAによる発現抑制により、細胞増殖が促進された。NLRR2の過剰発現は、Tunicamycin処理と同様にERマーカーであるPDI発現を誘導し、ERストレスマーカーであるCHOPおよびATF-4の発現を誘導した。これらは、CHOPとATF-4のluciferase reporterを用いた系においても発現誘導を確認できたことから、NLRR2によりCHOP,ATF-4の転写が促進されているものと思われた。したがって、NLRR2は小胞体ストレスの活性化を介して細胞死を誘導していることが明らかになった。(3)NLRR3は神経芽腫細胞の分化を促進したが、現在、ノックアウトマウス作成のためのコンストラクトが完成し、ES細胞作成の段階に入っている。
