一卵性双生児を対象とし後天的遺伝要因に着目した自己免疫疾患予後予測検査法の開発

Research Abstract

リンパ球サンプルの収集を行い、リンパ球をサブセット特異的に分離して、特定のサブセット由来のゲノムDNAを収集する方法を確立した。この方法を用いて、自己免疫性甲状腺疾患患者のサンプルとなるゲノムDNAを収集中である。
DNAのメチル化率解析については、メチル化特異的PCR法あるいはメチル化感受性制限酵素を用いたRFLP法によりメチル化検出法の基本的プロトコールの確立を試み、その結果、メチル化特異的PCR法ではサンプルが多量に必要であり再現性が充分でないことを見出した。そのため、メチル化感受性制限酵素を用いたRFLP法を中心に解析する必要があることがわかり、同法を用いたプロトコル作成中である。現在、やや荒い定量には再現性が認められる段階になった。
ゲノムDNA多型のうち、自己免疫性甲状腺疾患である橋本病の重症度やバセドウ病の難治性といった病態と関連して一卵性双生児での解析の基礎データとなるであろういくつかの多型を同定することに成功し、学術誌および国内・国際学会において発表した。
一卵性双生児の疾患表現型が異なるペアについては充分なサンプルが収集できていないため、未だこれらのサンプルについての明確な結果を示すには至っていない。引き続きサンプリング対象をスクリーニング中である。
