ガンマデルタT細胞と樹状細胞を用いた癌幹細胞特異的免疫療法の開発

Research Abstract

癌治療において、癌幹細胞を治療することができれば、再発・転移を含めて難治性癌をコントロールできる可能性がある。本研究では、癌幹細胞を標的とした新規治療モデルを作製し、治療抵抗性である癌に対する新たな治療戦略としての免疫療法の試みを行っている。
MH129細胞(C3Hマウス由来、肝細胞株)を用いて、癌幹細胞のみ、非癌幹細胞のみ、また同時投与したマウスに対する抗腫瘍効果解析を行った。細胞周期による細胞選択方法により、sub G1グループに癌幹細胞が豊富に含まれることが明らかとなった。本年度は、我々の研究より明らかになった新規肝癌細胞幹細胞マーカーCD13の発現の有無により癌幹細胞を抽出することが可能となった。それらは、肝臓細胞と胆管細胞という異なった種類の細胞に分化することを細胞表面マーカーの検索により明らかにした。癌幹細胞グループは未分化な細胞が発現する細胞表面マーカー(サイトケラチン14)も同時に発現した。また、宿主マウスに投与し、癌幹細胞グループがより少ない投与細胞数で腫瘍塊が生着、増殖することを明らかにした。
さらに、それらの癌幹細胞グループ細胞と親株細胞の遺伝子発現パターンを解析するために、それぞれマイクロアレイを行い、癌幹細胞に特異的に発現する遺伝子群を同定した。また、同様の手法により、癌幹細胞に特異的に発現が抑制される遺伝子群の同定を行った。
ガンマデルタT細胞をマウスリンパ球とゾレドロン酸との混合培養によって誘導を行った。ヒトガンマデルタT細胞を様々な条件の下で誘導条件を行い、最も有効でかつ効率よく分化誘導できる条件を決定した。今後は癌幹細胞特異的免疫応答の解析を行う。
