Outline of Final Research Achievements |
Microsomal prostaglandin E synthase (mPGES)-1 and prostacyclin synthase (PGIS) are terminal enzymes that produce PGE2 and PGI2, respectively, downstream of cyclooxygenase (COX). We analyzed role of mPGES-1 and PGIS in chemical-induced bladder diseases. In PGIS KO mice, cyclophosphamide-induced cystitis was significantly suppressed but not in mPGES-1 KO mice. On the other hand, N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN)-induced bladder carcinogenesis was suppressed in mPGES-1 KO mice. However, in PGIS KO mice, BBN-induced bladder carcinogenesis was promoted.

Academic Significance and Societal Importance of the Research Achievements |
化学物質曝露に伴う膀胱疾患の発症・進展に、PG類の産生が関わることが予想されるが、膀胱疾患の発症や進展とPG合成酵素との関連についてはほとんど解析が進んでいなかった。本研究によって化学物質による膀胱炎や膀胱がんの発症にmPGES-1およびPGISが関わることが示唆され、これらの疾患の治療標的となりうることが期待される。新たな作用機序の治療法となれば、治療の選択肢が増えることになり、社会的意義も大きいと考えられる。
