Outline of Annual Research Achievements

アレルギー疾患における難治化機序の解明とそれに対する治療戦略の構築は, 当該領域における重要な課題である。我々は，線維芽細胞から産生されるマトリセルラータンパク質であるペリオスチンが，さまざまな炎症疾患に関与するとともに, 炎症における線維化形成に深く関与していることを明らかにしてきた。これに基づき, ペリオスチンがアレルギー疾患における新規の創薬標的となるとともに, アレルギー病態を反映するバイオマーカーとなることを示してきた。本研究では，ペリオスチン研究をさらに発展させ，アレルギー疾患の難治化機序の解明とそれに対する治療戦略の構築の課題に対するプロトタイプ（原型）となる解答を示すことを目指している。
本研究における昨年度の代表的な研究成果として，以下があげられる。我々は，以前，顔面に限局した皮膚炎と激しい痒みによる引っ掻き行動を示すFADS（Facial Atopic Dermatitis with Scratching）マウスを確立した。生まれつきペリオスチン遺伝子を欠損するFADSマウスを作成したところ, 皮膚炎と引っ掻き行動が改善したことから, FADSマウスにおいてペリオスチンが皮膚炎と痒みの発症に重要であることを明らかにした（Nunomura, Cell Rep, 42, 111933, 2023）。さらに, 低分子化合物であるCP4715を投与すると皮膚炎と引っ掻き行動が改善したことから, CP4715はアトピー性皮膚炎に対する創薬となる可能性を示した。
この研究成果は，ペリオスチンを標的としたアトピー性皮膚炎に対する新規の創薬に応用できる点で, 非常に有用であると考えられる。
