Outline of Annual Research Achievements |
膵がんは罹患数と死亡数が近似する極めて治療成績不良ながんである。そのため、膵がんが発生する超初期段階の解析と、がんへの進行を食い止める方策は重要である。多くの先行研究および独自の検討により、膵発がんの超初期段階では腺房導管異形成(acinar-to-ductal metaplasia: ADM)とKras変異の併存が必要なことが示されている。ADMは炎症など微小環境の変化によって腺房細胞が脱分化した化生性変化であり、腺房細胞に再分化できるユニークな可塑性を持つ。本研究ではADMを中心に、細胞系譜イメージング技術と網羅的解析を用い、以下の検討を行った。 (1)Dclk1陽性ADM細胞からの細胞系譜解析が可能なDclk1CreER/+; Rosa26mTmG/+マウス、Dclk1CreER/+; Rosa26mTmG/+; Pdx1-Flp; KrasFSF-G12D/+マウス等に腹壁イメージング・ウインドウを設置し、膵炎によりADMの脱分化・再分化が生じる過程、およびDclk1陽性ADM細胞から膵腫瘍、さらに膵がんが生じる過程を二光子顕微鏡で経時的に観察した。 (2)Dclk1CreER/+; Rosa26mTmG/+マウスからADMの脱分化・再分化過程で腺房細胞、ADM細胞を経時的に収集し、ADM細胞に生じる動的なシグナル変化の網羅的解析を行った。それに基づき、シグナルの活性化・阻害実験を行い、脱分化・再分化に及ぼす影響を検証した。また、マウス腺房細胞に加え、ヒト腺房細胞にも遺伝子操作を加え、ADMまたはPanIN細胞に変化させる試みにも取り組んだ。 これらの検討を通じて、膵発がん超初期段階の本質に迫り、ADMの可塑性を活かして正常へ再分化させる=超初期段階から引き返す膵がん予防戦略の手がかりを得ることができた。
