電荷による細胞応答制御法の創出ーチャージハイドロゲルでウイルス感染症・がんを治す

Outline of Annual Research Achievements

(研究の骨格)医学の領域で感染症・がんは2大疾患であり、これらを制御することは喫緊の課題である。我々は最近、バイオマテリアルの1つであるハイドロ ゲルがヒト細胞に急速なゲノム変化を誘導することを見出した(Nat. Biomed. Eng, 5, 914-925, 2021)。本研究はこの技術を格段に発展させるもので、ハイドロゲルを用いて細胞周囲のチャージ(荷電状態)を変化させることで細胞のゲノム状態を制御し、1)ウイルスへの感染を防御し、また、2)がん幹細胞を誘導 することで、がんの早期診断法およびがん幹細胞特異的治療薬を開発する。
(今年度の実績)(1)チャージゲルによるウイルス増殖制御法の創出:陰性荷電を有するAMPSと陽性荷電を有するAPTMAモノマーを様々な割合で配合したチャージハイドロゲルを作製し、ウイルス感受性細胞VEROを培養し、COVID-19感染後にウイルス増殖を検討した。VERO細胞が効率よく接着し、ウイルス増殖が亢進するチャージゲルを見出した。 (2)チャージゲルによるがん幹細胞誘導:AMPSとAPTMAモノマー、および中性モノマーDMAを様々な割合で配合したチャージハイドロゲルを作製し、ヒト膠芽腫細胞株3種を培養して、癌幹細胞マーカーの発現が最も亢進するチャージの条件を見出した。最適化したチャージゲル上に癌細胞は接着し、特定の蛋白質の発現亢進(260 kDa, 55 kDa)および低下(45 kDa, 38 kDa)が認められ、これらの蛋白質発現変動が癌幹細胞の誘導に重要であることが明らかとなった。
以上より、ウイルス増殖およびがん幹細胞の創出において、細胞周囲の特定のチャージ(荷電状態)が重要であることが明らかとなった。これらをコントロールすることがウイルス感染およびがん幹細胞標的治療に重量であることが示唆された。
