Outline of Final Research Achievements |
Using Cre-loxP systems, we generated a new mouse model (fmSirt1KO mice) in which the Sirt1 function was selectively deleted in the fast-twitch muscle fibers. In this model, we analyzed the body weight and muscle mass, as well as muscle function and metabolic changes. In young mice, there was no difference in muscle mass or function. However, metabolic changes, including changes in dark-phase oxygen consumption, were suggested with aging.

Academic Significance and Societal Importance of the Research Achievements |
骨格筋におけるSirt1作用はミトコンドリア代謝などを介し, 筋質に影響をおよぼす一方, 筋重量維持には大きく影響しない可能性が示唆された. またSirt1機能は年齢依存的に変化する可能性が考えられた. 今後Sirt1アゴニストによる機能回復などを検討することで, 社会的課題であるサルコペニアの治療や予防の一助となる可能性が期待される.
