Outline of Final Research Achievements |
The ratio of 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) were measured for residents undergoing medical examinations. We examined whether 5mC and 5hmC in leukocytes were associated with lifestyle-related diseases. As a result, participant with a history of cardiovascular diseases had a high 5hmC levels in leukocytes. Therefore, we suggested that DNA demethylation could be associated with cardiovascular diseases in Japanese population.

Academic Significance and Societal Importance of the Research Achievements |
DNAメチル化及びDNA脱メチル化は、遺伝子配列に依存しない遺伝子発現の制御機構であり、様々な生活習慣病の発症と進展に関与していることが明らかとなっている。従来、DNAメチル化レベルと生活習慣病に焦点を当てた疫学研究が主体であり、DNA脱メチル化に焦点を当てた疫学研究は限定的である。DNAメチル化およびDNA脱メチル化の双方と生活習慣病との関連についてエビデンスを構築することは、発症メカニズム解明およびバイオマーカー確立に寄与することができると考えている。
