Outline of Annual Research Achievements

成長因子を固定化することで、細胞の成長や分化のような工事の細胞機能を制御できる新しいバイオマテリアルが生み出せることを明らかにしてきた。本研究では、進化分子工学の方法を用いて新しい結合性成長因子を調製し、医用材料としての展開をはかった。第一には、無細胞翻訳系を用いたディスプレイ法を用いたペプチド進化分子工学法の確立、第二には、非天然アミノ酸を導入して結合性を強化した成長因子を調製、第三には、これらの方法を用いた新しい機能性ペプチドの合成、そして第四には、調製した成長因子や機能性分子の医療応用を行った。
第一については、血管内皮細胞成長因子(VEGF)についてコラーゲン結合性を付与することに成功した。これはVEGFをコードするDNA配列に直接ランダム配列を導入し、コラーゲンに対するアフィニティ選別を行うことで遂行した。得られたVEGF誘導体は、コラーゲン結合性を持つとともに、血管内皮細胞の成長促進活性ももつことがわかり、従来知られているコラーゲン結合性ペプチドをそのまま導入した場合より高い活性を有していた。
第二については、い貝が分泌する水中接着タンパク質の活性部位である3,4-ジヒドロキシフェニルアラニン(DOPA)を含むペプチドをVEGFや骨形成タンパク質(BMP)へ連結させることに成功し、これらがクロム合金やチタンへ結合し、各々の生物活性を発揮することを明らかにした。
第三には、蛍光発生基、電気重合基、阻害剤、光異性基などを側鎖に導入した非天然アミノ酸を組み込んだ進化分子工学を可能にし、新しい機能性ペプチドの調製に成功した。
第四には、結合性成長因子の医療応用として、動物実験評価を行った。特にBMP誘導体が、損傷骨や脊椎の再生活性に有効であることを明らかにした。
