Research Abstract

コスタリカ共和国グアナカステ自然保護区サンタロサ地区において、ヒトと同様な恒常的3色型覚をするとされる新世界ザルのホエザルと2色型と3色型の多型的色覚を有する新世界ザルのオマキザル及びクモザルから糞サンプルを収集した。また、乾季においてオマキザルの採食する果実を収集した。糞サンプルからはDNAを抽出し、赤-緑オプシン遺伝子のエクソン3及び5のPCR増幅と一部の塩基配列決定を行なった。味覚受容体の味覚受容体TAS2R1、TAS2R4、TAS2R40についてシロガオオマキザル4群、チュウベイクモザル1群、マントホエザル4群、グアテマラホエザル3群に対し総計1144run(約0.6Mb)の集団塩基配列データを収集した。ホエザルの集団サンプルからは赤-緑オプシンのエクソン5において赤型と緑型のハイブリッド型を見いだし、恒常的3色型色覚とされるホエザルに色覚多型がある可能性を示した。一方、本研究のサンプルの大部分が糞DNAであることの利点を活かし、本研究と密接に関係する野生霊長類の食生態の解明に資するために、腸内細菌叢の分類同定を16SリボソーマルRNA遺伝子を標的として行なうことにした。ホエザルの糞サンプルを用いて腸内細菌叢の解析に向けて、糞DNAの保存法、抽出精製法等の実験条件の検討のためのパイロットスタディとして4サンプルの検討を行なった。さらに今後フィールドサンプリングでは回避し難い土壌や降下物の混入や昆虫接触の影響を評価するパイロットスタディが必要とされる。
