| Research Abstract |
メガネザル3種（ボルネオメガネザルTarsius bancanus、フィリピンメガネザルT. syrichta、スラウェシメガネザルT. tarsier）のL/Mオプシン遺伝子を単離した。近縁なボルネオメガネザルとフィリピンメガネザルはそれぞれMタイプ、Lタイプと異なるのに対し、それらの外系統となるスラウェシメガネザルはLタイプであった。イントロンとエクソン同義塩基サイトによる遺伝子系統関係は種の系統関係と一致したのに対し、エクソン非同義塩基サイトによる遺伝子系統樹では同じLタイプのフィリピンメガネザルとスラウェシメガネザルがより近縁でMタイプのボルネオメガネザルは外系統となった。このことはそれら3種の共通祖先でLとMタイプが種内多型として存在したことを支持しており、したがって新世界ザルと類似して3色型色覚が一部のメスに現れる多型色覚であったことを支持している。さらにこのことはそれらが昼行性あるいは昼夜行性であったことを支持している。一方、メガネザルの共通祖先は化石証拠からすでに巨眼であったことが知られており、このことはそれらが夜行性であったことを支持している。一見相矛盾するこれらの結果は3色型色覚が昼行性になってから進化したのではなく、霊長類の祖先が完全な夜行性活動性でなく、桿体と錐体の両方が機能しうる薄明時活動性であり、そこで3色型色覚も進化した可能性を示唆している。

Reason
主要な研究対象と予定していた新世界ザルに加え、原猿類であるメガネザルの祖先種に3色型色覚を含む色覚多型があった可能性を示すことができた。新世界ザルであるオマキザルとクモザルの苦味受容体遺伝子TAS2R1について、それぞれのアミノ酸レベルの多型に対し受容体再構成を行ないカルシウムイメージングを実施した。確定的結果は得られていないが、ヒトで示された苦味物質に対する感受性がヒトとは異なることが示唆された。

| Strategy for Future Research Activity |
色覚の様相の異なるホエザル（恒常的3色型）、オマキザル（3アレル型多型色覚）、クモザル（2アレル型多型色覚）、ヨザル（1色型完全色盲）に対し、苦味受容体のin vitro機能解析を進める。ホエザルに見出された3色型色覚の多型及びメキシコのクモザルに見出した新規L/Mオプシンアレルの頻度調査と視物質再構成実験を進め、非狭鼻猿類の色覚多型と狭鼻猿類の恒常的3色型色覚の進化的意義についての理解を進める。
