Research Abstract

交付申請書に記した研究実施計画の項目ごとに報告する。(１）赤オプシン（LWS, 552 nm）、第二の緑オプシン（RH2-C, 490 nm）、第二の青オプシン（SWS2B, 416 nm）、および紫外線オプシン（SWS1, 367 nm）について吸収極大を明らかにした。第三の緑オプシン（RH2-A）は偽遺伝子であることが判明した。 (２）発光ダイオード光（赤、緑、青）がマツカワの視床下部-脳下垂体系のホルモン遺伝子に及ぼす効果を調べた。［視床下部］プロオピオメラノコルチン（POMC）遺伝子の発現が青および緑色光により上昇した。メラニン凝集ホルモン（MCH）遺伝子の発現は青および緑色光照射により低下した。オレキシンおよび神経ペプチドＹ遺伝子の発現に変化は認められなかった。［脳下垂体］POMC遺伝子の発現が、視床下部での動態と同様に青および緑色光照射により上昇した。成長ホルモン遺伝子発現は緑色光下において減少傾向が認められた。 (３）青色光がキンギョにおいてMCH遺伝子の発現を促進することを再現できた。緑および赤色光では上昇しない。また同一の試料において、POMC遺伝子の発現は緑および赤色光で上昇し、青色光では変化がなかった。 (４）昨年度マツカワにおいて第二のMCHを発見した。今年度はその一次構造を明らかにした。また、質量分析により当該ペプチドを同定した。本ホルモンの活性および背地色に対する応答は第一のMCHと同等であることを認めた。 (５）体色調節に関与する黒色素胞刺激ホルモンをコードする、第二および第三のPOMC遺伝子を発見した。第二POMCは脳と脳下垂体で発現することから、既知の第一POMCと同様の機能を有すると考えられる。第三POMCは脳下垂体と頭腎で発現することから、未知の生命過程に関与するものと推測される。
