| Research Abstract |
本研究では現在頭頸部癌の検体を用いた遺伝子解析が進行中である。先んじて行ったトランスクリプトームによる解析では、多くの検体がEGFRパスウエイ関連遺伝子の増幅を認めている。またEGFRが低発現の群にあっては他のパスウエイがそれを相補するように上昇していることが分かった。そのうちの一つであるIGFIRパスウエイは上皮性腫瘍の成長因子としてEGFRパスウエイと下流が重複し、EGFRを相補する働きがある事を頭頸部癌細胞株を用いたin vivoの実験でも同様の結果が認められている。また間葉系のパスウエイもNF-KBパスウエイやFGFRパスウエイの相補効果があることが遺伝子解析の結果から示唆されている。今後にはこれらのパスウエイの遺伝子に過剰発現がみられるそれぞれの頭頚部癌細胞株を用いて、リガンドや阻害剤の有無による下流の活性化の変化を追う事で、メカニズムの解明にあたりたい。これらのパスウエイはこれまでに他癌においてそのパスウエイが癌細胞の増殖、転移に関連していることが示されており、すでにそれぞれの分子標的治療薬が開発されてきている。頭頸部癌でもこれらのパスウエイを阻害することで癌増殖抑制効果があることが見込まれる。今後はさらにパスウエイのスイッチングによる分子標的治療の耐性化のメカニズムの解明も期待している。また平行して正常部位におけるパスウエイの影響を分析し、副作用の予測を検討していく。この研究結果がより一層の頭頸部癌治療の予後改善へのブレイクスルーとなることを期待する。

現在は頭頸部癌細胞株による研究を主に行なっており、トランスクリプトーム解析ではこれまでに報告された様々ながん遺伝子が頭頸部癌においても他癌同様に発現が変化していることがわかった。臨床検体でのトランスクリプトーム解析およびマイクロRNAの網羅的解析併せても進んできている。

| Strategy for Future Research Activity |
データの蓄積を待ちつつ、シグナル伝達における各分子の役割、書く分子相互のクロストークの解析を行り。新たな知見に関してはウエットな実験による検証および臨床検体での検討が必要である。今後は当該分子のトランスフェクションやノックダウンによる癌細胞の増殖シグナルの増減を確認し、実際の分子標的薬による耐性化のメカニズムについても検討を重ねる。
