Research Abstract

TNF-α・Claudin-1経路により制御される遺伝子を解明するため、TNF-α処理の有無別、Claudin-1 siRNA導入の有無別にMKN28をサンプリングし、DNAマイクロアレイによる網羅的解析を試みた。TNF-α処理によりmRNA発現レベルが変化した遺伝子は、control/ Claudin-1 siRNA導入細胞で932/789種であった。両者を詳細に解析すると、TNF-αにより発現変化した遺伝子のうち約70%が、Claudin-1のノックダウンにより抑制されることが明らかとなった。また、同条件でmicroRNAマイクロアレイを行ったところ、TNF-α処理により変化したmicroRNAは、control/ Claudin-1 siRNA導入細胞で378/191種であった。TNF-αにより発現変化したmicroRNAのうち約87%が、Claudin-1のノックダウンにより抑制された。これらDNA・microRNAマイクロアレイの結果より、TNF-αシグナル伝達におけるメディエーターとしてのClaudin-1の機能を解明するとともに、同経路により多くのmRNA・microRNA発現が制御されていることが示された。また、Claudin-1 siRNA導入細胞における、細胞増殖・細胞浸潤抑制効果、アポトーシス増強効果を新たに見出した。DNAマイクロアレイの結果をPathway解析したところ、MMP7、TGFBR1、TNFSF10を含む経路がClaudin-1に関連したトップネットワークの一つとして抽出された。これらの成果は既に学会で発表し英文論文としてまとめている。また同時に、肺癌細胞株を用いた基礎的解析を行い、Claudin-1がTNF-αによる腫瘍活性化シグナルを伝達するメディエーターとして機能することを明らかにした (PLoS ONE. 2012）。
