Outline of Annual Research Achievements

本課題は、申請者が従来取り組んできた炎症性腸疾患の難治性潰瘍を腸上皮幹細胞オルガノイドで被覆し、その完全な治癒を目指す細胞治療を推進する研究を基に、腸から樹立したオルガノイドを自在にデザインし、肝不全・糖尿病などの全身的代謝疾患の治療・予防に有効な新しいデザイン化オルガノイド『ArtDO』(Artificial Designed Organoids)を開発し、それを腸へ移植することで異所性の機能性臓器の個体内での創出を目指すArtDO構想を推進し、全身性代謝異常やホルモンネトワーク異常の是正による代謝疾患の新しい治療措置の樹立を目指す課題である。
研究初年度に当たる2022年度には、腸から樹立したオルガノイドを、肝細胞と親和性の高い吸収上皮系の細胞に誘導する方法を確立した。同時に、吸収上皮細胞から肝細胞への誘導に関して、これをブロックする腸上皮特異的な転写因子の存在も明らかにしている。また現在、腸内分泌細胞に膵臓が有するホルモン分泌能を導入することを目指して、まずは重要な膵内分泌ホルモンであるインスリン分泌機構に着目し、インスリン分泌を制御する責任遺伝子の同定に取り組んでいる。なお、ArtDO-Pancreasにおいては、手術検体、EUS-FNA検体から膵GEP-NENオルガノイドを樹立し、他疾患も合わせて幅広く膵臓オルガノイドバンクを予定通り樹立している。同時にRNAの採取を行いRNAーseqを行う準備も行っている。インスリノーマ由来オルガノイドはインスリンを分泌することを確認済みであり、予定どおり解析に用いる。

Reason

腸上皮細胞を肝細胞に誘導するArtDO-liverにおいても、腸上皮細胞を膵島b細胞へ誘導するArtDO-Pancreasにおいても、研究計画に記載した通りに実験が進捗しており、概ね計画通りに進行している。

Strategy for Future Research Activity

ArtDO-liverにおいては、2022年度に同定した肝細胞への成熟を阻害する、腸らしさを頑強に維持する転写因子を阻害する方法を、主に遺伝子操作で行うよう進めていく。ArtDO-Pancreasにおいては、RNAシーケンスなどの手法で、樹立したバイオバンクの解析を進めていく予定である。
