Outline of Annual Research Achievements |
The establishments of the UPLC-MS system and the Benchtop NMR system have enabled the scale-up production of the essential chemicals. Various methods for the UPLC-MS system and the Benchtop NMR system have been generated and optimised. Large-scale expression of the target protein has also been achieved. The protocols for large-scale affinity chromatography, ion-exchange chromatography and size-exclusion chromatography with the AKTAgo FPLC system have been extensively explored and optimised. SDS-PAGE characterisation of the fractions from each chromatography purification gave the desired purity in good quantity. Thus, scale-up protein production and purification have also been achieved. The subsequent scale-up production of nanopores and follow-up purification is in progress. Various methods are being examined for the best results. Initial measurements with the relevant biophysical instruments are encouraging. Hopefully, desired nanopores with very high purity would be obtained soon so that the desired properties could be achieved.
