Outline of Final Research Achievements

In this study, we aimed to elucidate the mechanism of transcription regulation by MED26, one of subunits of Mediator complex, through its nuclear droplet formation. The results of this study showed that MED26 interacts with various RNAs, that nascent RNAs are required for MED26 droplet formation, and that MED26 droplet formation is dependent on its intrinsically disordered region. These results of this research are expected to lead to the elucidation of a novel mechanism of transcription regulation by Mediator complex.

Academic Significance and Societal Importance of the Research Achievements

メディエーター複合体は遺伝子発現制御において中心的な働きをする巨大な転写制御複合体であり、近年、その液滴形成を介した転写制御機構が世界的に注目を集めている。本研究では、その機能的サブユニットであるMED26に着目し、MED26タンパク質の液滴形成能についての解析を行うことで、メディエーター複合体の転写制御機構を解明しようと試みた。本研究によってMED26の液滴形成についての新規の知見が得られたことで、メディエーター複合体を中心とした未知の転写制御機構の解明につながると期待される。
