Outline of Final Research Achievements

There were no significant differences in patient backgrounds between the 8 symptomatic and 12 asymptomatic groups and the 8 stable (S) and 12 unstable (U) plaque groups, respectively.
Carotid plaque specimens were prepared and subjected to HE staining and immunostaining for MPO, Histone H3, and PAD4. In the inflammatory cell accumulation sites, neutrophils were rarely observed, and monocytes and macrophages were predominant. Immunostaining showed no significant difference in the number of MPO-, Histone H3-, and PAD-positive cells between the each two groups.

Academic Significance and Societal Importance of the Research Achievements

頚動脈狭窄症に対して内膜剥離術を施行した患者を対象に、頚動脈狭窄症の発症様式やプラーク性状の違いに好中球タンパクシトルリン化がどのように関与しているかを検討した。症候性群・無症候性群、並びに安定プラーク群・不安定プラーク群の2群間で患者背景に違いはなく、プラーク内の活性化好中球数やヒストンH3、PAD4陽性細胞数にも違いはなかった。これまでの研究では好中球シトルリン化は細胞接着現象に関与していることが示唆されているが、本研究結果からは頚動脈狭窄症の発症形式やプラーク性状の違いには関与していない可能性が示唆された。
