維管束幹細胞の発生運命制御機構の解明

Outline of Annual Research Achievements

本研究の目的は、植物成長の鍵となる植物メリステムの組織構築機構を、幹細胞の発生運命制御の解明を通して明らかにすることにある。今年度は、TDIF機能を中心に維管束幹細胞の維持機構と、維管束幹細胞の成立機構、さらには維管束幹細胞からの木部細胞分化機構を、以下に示す3つの観点から解析した。

1.WOX4の下流因子の解明: WOX4ターゲット候補遺伝子について、その機能をT-DNA挿入変異体を用いて解析したが、顕著な表現型は得られなかった。そこで、更なるターゲット遺伝子の探索のために、チップシークエンスを行うこととし、その準備を行った。
2.ブラシノステロイドシグナル伝達とTDIFシグナル伝達のクロストークの解析: ブラシノステロイドシグナルとTDIFがGSK3-BES1経路を介してクロストークすることが明らかになったので、これらの2つのシグナル系がどのようにGSK3-BES1経路の制御を行っているのかを、BES1ファミリーを中心に解析した。その結果、2重変異体の作出に成功した。
3.leaf disk培養を用いた維管束幹細胞成立の解析: 私たちが昨年までに開発したleaf disk培養系では、葉肉細胞から維管束幹細胞の分化を経て、木部細胞・篩部細胞が分化する。そこで、この実験系を用いて、マイクロアレー解析を行い、葉肉細胞から維管束幹細胞への分化に関わる新規因子の同定を行った。その結果、多くの新規の遺伝子が同定され、そのうちには、NACタイプの転写因子も含まれていた。また、維管束細胞成立の研究において、新規のシグナル伝達分子の同定に成功した。

Strategy for Future Research Activity

今後、維管束幹細胞の維持機構、維管束幹細胞の成立機構、維管束幹細胞からの木部細胞分化機構の解明を、特にleaf disk培養を中心にして進め、そのシグナル伝達のアウトラインを明らかにする。
