次世代シーケンサーによる老年病の新規ホルモン作用標的の解明と抗加齢医学への応用

Research Abstract

本研究は個体と臓器の老化・老年病におけるホルモンの分子作用メカニズムを、次世代シーケンサーを活用して解明し、新しいシグナル経路とそれらのクロストーク、新規標的因子、共役因子、修飾因子とその役割を明らかにし、臨床応用を目指すものである。基礎医学、臨床医学、生物学の手法を活かし、新しい臨床学問領域、抗加齢医学への応用を目指す。
(1) 老化と老年病に関わる新規ホルモン受容体標的因子の同定、解析:老化と老年病に関連する核内受容体標的因子に関して、分子機能解析と生体機能解析を進め、老化と老年病における役割を明らかにし、老年病治療、抗加齢の標的として検討を加えた。そのため、次世代シーケンサーを活用し、正常及び老化細胞、老化モデル動物、老化組織やホルモン受容体関連遺伝子改変動物より、14-3-3zetaをはじめとする新規核内受容体標的因子の同定を進めた。
(2) ホルモン受容体新規シグナル経路とそのクロストークの解明と老化、老年病における意義:抗加齢、抗老年病作用に関わるホルモン受容体に対する転写活性、作用機序の差異を調べ、あわせて共役因子との相互作用、蛋白修飾について解析した。転写調節因子FOXファミリーと核内受容体の新しい相互作用を明らかにした。
(3) 新規ホルモン受容体シグナル経路、標的因子の機能・ゲノム解析と抗加齢医学への応用:ホルモン受容体関連因子の遺伝子改変動物、また新規開発した老化・老化疾患モデル動物を用いて、骨、血管、脳神経系をはじめとする各組織における老化ならびに老年病表現型を調べた。特に、ERシグナルを軟骨で増大させたマウスは骨格異常を示すなど治療薬開発のモデル動物を開発した。

Strategy for Future Research Activity

今後とも、核内受容体と老化・老年病の関連を抗加齢医学、老年医学、ゲノム医学、分子細胞生物学、生化学の手法を活用して多面的な観点から研究を進めていく。
