| Research Abstract |
1.平成23年度に得られた成果の発表（長嶋，秋本）：前立腺癌ではすでにaPKCλ/ιが高発現し，AP-1, NFκBからIL6を誘導し，アンドロゲン受容体が高発現することで，アンドロゲンが小量であってもこれを克服することを示した．実際に手術的に採取された前立腺組織では予後不良な症例で、aPKCλ/ιとIL6が正相関して高発現することを示し論文報告を行った（Ishiguro, Akimoto, Nagashima et al., Cancer Sci, 2012）。膵癌、メラノーマに関しては論文投稿、子宮頸癌と前癌病変、および悪性中皮腫については今年度、検討を進める予定である。2.aPKCλ/ιを分子標的とした乳癌，前立腺癌治療モデルの確立（秋本，長嶋，石黒，上村）：すでに乳癌でaPKCλ/ιが高発現し，oncogenicに働いている可能性を報告している（Kojima, Akimoto, Nagashima et al.Hum Pathol, 2008）．乳癌および前立腺癌でaPKCλ/ιを高発現している細胞株に対し，siRNA法やaPKC阻害薬(aurothiomalate)を用いて増殖抑制が得られるかを検討する．示標として，増殖曲線，軟寒天板上でのコロニー形成性，血清依存性，ヌードマウスでの造腫瘍性の変化などを見る．またIL6阻害薬（tocilizmab）の効果あるいは併用効果も検討し，aPKCλ/ι-IL6系の治療標的としての可能性を検証する．3.aPKCλ/ιとPAR3, PAR6との結合，共局在の検討（長嶋，秋本，石黒）：平成23年度に計画していた本項目は、人体材料を用いた研究が先行して進捗したため、および研究目的にかなった抗体が入手できなかったため、本年度に実施することとした。

Reason
当初、予定していた膵癌、悪性黒色腫におけるaPKCλ/ιの高発現が予後不良因子であることの論文発表については現在修正再投稿中である。細胞を用いたaPKC阻害剤を用いた腫瘍増殖抑制実験は適当な細胞株がなく、現在、検索中であるとともに、臨床検体から実験に適した細胞株の樹立を試みている。

| Strategy for Future Research Activity |
1.現在修正投稿中の膵癌、悪性黒色腫についての論文発表を完了する。2.後発で進めている悪性中皮腫、膀胱癌について倫理委員会に追加申請を行い、承認後、臨床検体を収集、研究を進める。3.細胞を用いた実験については目的に合致したaPKC高発現前立腺細胞株などを入手でき次第推進する。

細胞培養と動物実験、免疫組織染色に必要な試薬の購入及び論文校正費を予定している。また、研究成果発表としての学会参加費、論文掲載費も予定している。
