酸化ストレス誘導性の肝腫瘍原性遺伝子に関する機能解析：分子標的治療への基礎的検討

Outline of Annual Research Achievements

肝発癌は様々な原因による持続的肝傷害と密接に関連しているが、分子生物学的な発癌過程は原因により異なり、種々の遺伝子が複雑に相互関連している。しかし、その共通の病態は長期の炎症状態と酸化ストレスへの持続的曝露であり、酸化ストレス誘導性遺伝子群の中に多様性のある多段階的発癌に最も共通して関与する肝腫瘍原性遺伝子がある可能性は高い。本研究の目的は、申請者が報告してきたIQGAP1やvimentinの肝腫瘍原性遺伝子としての機能解析を行うことである。
種々のヒト肝癌細胞株におけるmRNA/蛋白発現状況を明らかにした。遊走能・浸潤能のある細胞株に対するIQGAP1のknockdownは、その活動能が著しく低下する細胞株とそれ程低下しない細胞株に分かれた。IQGAP1高発現株は、より低分化で遊走能・浸潤能が高く、その中心的役割をIQGAP1が担っていると考えられた。Vimentinも異なる細胞株で中心的役割を果たしていることが示され、遊走能・浸潤能の低下には違った遺伝子をknockdownする必要性があると考えられた。高発現細胞株でknockdownにより変動する関連遺伝子についてmRNA/miRNAの網羅的解析を行い、興味深い所見が得られた（登録・投稿中）。
Putative miRNA targetsが逆相関している遺伝子に関して、遊走能・浸潤能に関与する細胞内遺伝子の動態解析、特にmRNAとmiRNAの相互関連性と調整機構について検討した結果、腫瘍促進性のIQGAP1が抑制的に作用する可能性も示唆され、細胞依存性やpartner遺伝子依存性に両面の性質をもつ可能性が明らかになった。細胞機能や細胞形態への影響と癌細胞の悪性度や抗癌剤への感受性との関連性についても今後の課題である。
