好酸球性中耳炎における炎症因子ネットワークの解明と新規治療戦略の確立

Research Abstract

ヒト好酸球性中耳炎の症例より中耳貯留液を採取し、サイトカイン・アレイ法を用いて貯留液中の炎症性タンパクを網羅的に解析した。その結果、好酸球性中耳炎症例の中耳貯留液中にはInterleukin (IL)-1β、IL-1ra、IL-1α、IL-2、IL-3、IL-4、IL-5、IL-6、IL-7、IL-10、IL-13、IL-15、IL-16、Angiogenin、PARC、monocyte chemotactic and activating factor、macrophage inflammatory protein -3α、neutrophil-activating protein-2、macrophage inflammatory protein -1δ、Insulin-like growth factor binding protein-1、Macrophage-derived chemokine、Eotaxin-2、Tumor Necrosis Factor-α、Tumor Necrosis Factor -β、Leptin、Bone morphogenetic protein 4、Monocyte chemotactic protein 2、thymus and activation-regulated chemokine、Macrophage-colony stimulating factor、Fit-3 Ligand、stem cell factor、Ciliary Neurotrophic Factor、Fractalkine、Granulocyte Macrophage colony-stimulating Factor、Insulin-like growth factor Iなど、多数の炎症性タンパクが有意なレベルで存在することが明らかとなった。

ヒト好酸球性中耳炎症例より順調に中耳貯留液のサンプルの採取が進み、その中の炎症性タンパクの網羅的検出を行うことができたため。

Strategy for Future Research Activity

予定通り、今後は各炎症性タンパクと臨床因子との関連を探索するとともに、動物実験モデルの作製を目指す。

抗体や各種炎症因子検出材料を購入するとともに、学会や研究会に参加し、成果を発表と情報収集を行う。
