Outline of Annual Research Achievements |
平成27年度における研究実施により、以下の成果が得られた。 海馬におけるAMPARの標的選択的制御について、シャーファー側枝が標的とする錐体細胞(有孔シナプスと無孔シナプス)、パルブアルブミン陽性介在ニューロン(PVシナプス)、パルブアルブミン陰性ニューロン(nonPVシナプス)のAMPARの発現密度を比較した。その結果、AMPARの発現密度は有孔シナプス>無孔シナプス、PVシナプス>nonPVシナプスの順に標的選択性が観察された。この発現密度の違いは、AMPARの調節分子であるTARPg-2とg-8のシナプス発現密度と相関していた。さらに、これらのTARPを欠損するマウスでは、これらのシャーファー側枝シナプスのAMPARの発現密度は均一となり、標的選択性が失われた。以上の結果は、TARPg-2とg-8が海馬CA1におけるAMAPRの標的選択性を決定する分子であることを示し、これらの研究成果はJournal of Neuroscienceに受理された。 黒質線条体ドパミン投射系の伝達様式を追求した所、このドパミンシナプスのシナプス後部にGABA受容体が発現する伝達物資と受容体がミスマッチした異種結合であることを発見した。ドパミンシナプスは,GABAシナプス選択的な接着分子ニューロリギン2が存在し,この分子を培養細胞で発現させると黒質ドパミンニューロンとの接触部位がドパミンシナプスへと分化した。反対に,中型有棘ニューロンでこの分子発現を抑制すると,ドパミンシナプスは減少した。以上の結果は,中型有棘ニューロンはGABAシナプスの接着分子であるニューロリギン2を利用してドパミンシナプス形成を促進し,このニューロンに対するドパミンによる調節の選択性や効率性を賦与していることを示唆する。この研究成果はProceedings of the National Academy of Sciences of the United States of Americaの電子版に2016年3月に公表された。
