Outline of Annual Research Achievements

①タンパク質分子の機能動態撮影:最終年度にあたるH28年度はこれまでに行ってきたタンパク質分子の機能動態撮影研究をまとめるためにデータの補強、データの解析を行い、論文として成果をまとめることに努力した。Nat. Nanotechnol.に投稿した天然変性領域の一般則を発見した論文を改定提出し、最終受理の報告を待っている段階である。酸化還元酵素の成果はScience誌に投稿した。Rett症に関係するMeCP2の成果はScienceに投稿すべく原稿執筆中である。その他に、Myosin Vと膜孔形成毒素の成果についても論文にまとめている。このように今後1年以内に優れた成果が続々と論文として公表される予定である。これらに加え、欧州の研究者と共著でAFMの分子・細胞生物学への応用に関する総説をNature Nanotechnology誌に発表した。
②高速AFMの機能拡張に欠かせない探針走査型高速AFMを完成させた。これまで問題となってきたZ走査に伴うカンチレバー振幅計測の誤差発生の問題を解決したことが大きい。細胞観察可能な広域・高速スキャナーの開発成果とともに、Ultramicroscopy誌に成果を発表した。本研究の成果を更に発展させる研究については、他のプロジェクトで進めている。
③高速SICMの開発:開発した要素技術(高速・低ノイズのイオン伝導計測器、ガラスキャピラリー走査用の高速スキャナー等)を組み込んだ高速SICM装置全体を組み上げ、性能試験を行った。イメージング速度は従来の100倍強向上し、目標を達成したと言える。実際、細胞動態、脂質膜の動態などが撮影された。この成果を公表すべく現在2本の論文を執筆中である。本研究では取り上げなかった高速SICMの空間分解能の向上は本研究終了後、別のプロジェクトとして本格的に進める計画である。

以上のように、本研究は当初の目標を十分に達成することができた。
